Baicalein restrains proliferation, migration, and invasion of human malignant melanoma cells by down-regulating colon cancer associated transcript-1 BJMBR
Yang,Xiaoliang; Jiang,Jinjie; Zhang,Chunyan; Li,Yinghao.
Baicalein (BAI) is an acknowledged flavonoids compound, which is regarded as a useful therapeutic pharmaceutical for numerous cancers. However, its involvement in melanoma is largely unknown. This study aimed to examine the anti-melanoma function of BAI and unraveled the regulatory mechanism involved. A375 and SK-MEL-28 were treated with BAI for 24 h. Then, CCK-8 assay, flow cytometry, and transwell assay were carried out to investigate cell growth, migration, and invasion. RT-qPCR was applied to detect the expression of colon cancer associated transcript-1 (CCAT1) in melanoma tissues and cells. The functions of CCAT1 in melanoma cells were also evaluated. Western blot was utilized to appraise Wnt/β-catenin or MEK/ERK pathways. BAI restrained cell...

Effect of Wnt/β-catenin and NF-κB signaling pathways on mucus secretion with hypertonicity in 16HBE cells BABT
Xiaoyan,Liu; Xiangdong,Zhou.
This study aimed at identifying the molecular mechanisms and effects of hypertonicity on mucin5AC (MUC5AC) expression in airway epithelial cells for which immortalized human bronchial epithelial (16HBE) cells were cultured in 600 mOsm/L hypertonic medium for different times in vitro. Proteins of MUC5AC and β-catenin, Cyclin D1, NF-κB p65 were detected by enzyme linked immunosorbent assay (ELISA), reverse transcription (RT)-PCR and western blotting assays. After transfection of β-catenin siRNA in 16HBE cells, the levels of β-catenin, Cyclin D1, NF-κB p65 and MUC5AC protein were detected. Results showed that the levels of mRNAs and proteins of target genes increased significantly after the exposure to hypertonic conditions and the expression content...

Glycoprotein nonmetastatic melanoma protein B accelerates tumorigenesis of cervical cancer in vitro by regulating the Wnt/β-catenin pathway BJMBR
Xu,Shuxiang; Fan,Yingying; Li,Dongping; Liu,Yan; Chen,Xu.
Cervical cancer is one of the most common cancers among women around the world. However, the underlying mechanism involved in cervical cancer progression is incompletely known. In the present study, we determined the role of glycoprotein nonmetastatic melanoma protein B (GPNMB) in tumorigenesis of cervical cancer. According to the GEO database, we found that GPNMB expression was significantly higher in cervical cancer than in normal cervix epithelium. A similar pattern was observed in GPNMB expression in cultured cervical cancer cells and normal cervical epithelial cells. Compared with the control, GPNMB knockdown significantly decreased the proliferation and migration capacity, but enhanced the apoptosis capacity of SiHa and HeLa cells. Additionally, the...

Propofol inhibits proliferation, migration, and invasion but promotes apoptosis by regulation of Sox4 in endometrial cancer cells BJMBR
Du,Qing; Liu,Jia; Zhang,Xuezhi; Zhang,Xin; Zhu,He; Wei,Ming; Wang,Shilei.
Propofol is an intravenous sedative hypnotic agent of which the growth-inhibitory effect has been reported on various cancers. However, the roles of propofol in endometrial cancer (EC) remain unclear. This study aimed to explore the effects of propofol on EC in vitro and in vivo. Different concentrations of propofol were used to treat Ishikawa cells. Colony number, cell viability, cell cycle, apoptosis, migration, and invasion were analyzed by colony formation, MTT, flow cytometry, and Transwell assays. In addition, the pcDNA3.1-Sox4 and Sox4 siRNA plasmids were transfected into Ishikawa cells to explore the relationship between propofol and Sox4 in EC cell proliferation. Tumor weight in vivo was measured by xenograft tumor model assay. Protein levels of...
